An unbroken cold chain is an uninterrupted series of storage and distribution activities which maintain a given temperature range. It is used to help extend and ensure the shelf life of products such as fresh agricultural produce, seafood, frozen food, photographic film, chemicals, and pharmaceutical drugs. Such products, during transport and when in transient storage, are called cool cargo. Unlike other goods or merchandise, cold chain goods are perishable and always en route towards end use or destination, even when held temporarily in cold stores and hence commonly referred to as cargo during its entire logistics cycle. Cold chains are common in the food and pharmaceutical industries and also in some chemical shipments. One common temperature range for a cold chain in pharmaceutical industries is 2 to 8 °C (36 to 46 °F). but the specific temperature (and time at temperature) tolerances depend on the actual product being shipped. Unique to fresh produce cargoes, the cold chain requires to additionally maintain product specific environment parameters which include air quality levels (carbon dioxide, oxygen, humidity and others), which makes this the most complicated cold chain to operate. This is important in the supply of vaccines to distant clinics in hot climates served by poorly developed transport networks. Disruption of a cold chain due to war may produce consequences similar to the smallpox outbreaks in the Philippines during the Spanish–American War. There have been numerous events where vaccines have been shipped to third world countries with little to no cold chain infrastructure (Sub-Sahara Africa) where the vaccines were inactivated due to excess exposure to heat. Patients that thought they were being immunized, in reality were put at greater risk due to the inactivated vaccines they received. Thus great attention is now being paid to the entire cold chain distribution process to ensure that simple diseases can eventually be eradicated from society. Traditionally all historical stability data developed for vaccines was based on the temperature range of 2–8 °C (36–46 °F). With recent development of biological products by former vaccine developers, biologics has fallen into the same category of storage at 2–8 °C (36–46 °F) due to the nature of the products and the lack of testing these products at wider storage conditions. The cold chain distribution process is an extension of the good manufacturing practice (GMP) environment that all drugs and biological products are required to adhere to, enforced by the various health regulatory bodies. As such, the distribution process must be validated to ensure that there is no negative impact to the safety, efficacy or quality of the drug substance. The GMP environment requires that all processes that might impact the safety, efficacy or quality of the drug substance must be validated, including storage and distribution of the drug substance.

India is an agricultural-based economy and is the largest producer of fruits and vegetables in the world. Fruits & vegetables, being perishable in nature require certain techniques of preservation for retaining the quality and extend the self-life of the production. The estimated annual production of fruits and vegetables in the country is about 130 million tonnes. The cold storage & cold chain facilities are the prime infrastructural component for such perishable commodities. Cold storage is a temperature – controlled supply chain network, with storage and distribution activities carried out in a manner such that the temperature of a product is maintained in a specified range, needed to keep it fresh and edible for a much longer period than in normal ambient conditions. A cold chain can be managed by a quality management system generally called as warehouse management. India’s warehousing requirement is expected to grow at an annual average rate of 9%. Natura Brazil Cosmetics is Brazil's biggest company for cosmetics, toiletries, perfumes and drugstore articles. It provides over 100 million customers in Latin America and Europe with its products. In order to be able to guarantee the high degree of customer satisfaction and to also reduce errors within the logistics chain, Natura assigned the general contractor SSI Schaefer for the planning and implementation of a logistics hub in Itupeva, São Paulo (Brazil). This distribution centre, with a storage area of 35,000 m2 and a capacity to store 3.6 million bins, is the biggest hub within the Natura logistics network. It provides two further hubs, all international companies as well as twelve distribution centres with Natura products. Twenty truck loads are handled daily thanks to an automated unloading system for trucks which was especially designed for Natura. This automated unloading system can transport 48 pallets simultaneously and unload them in less than 5 minutes fully automatically. In incoming goods, all pallets and loading units are checked for conformity at an inspection station before being stored in a vertical warehouse with 13 storage-retrieval machines of the type Exyz. The storage and retrieval process at the 90,000 pallet locations is extremely energy-efficient. Retrieval is handled by the flexible multi-level shuttle system Navette, which consists of twenty individual warehouse shuttles. The system organises more than 1000 different Natura products directly in the outgoing goods and depalletisation areas. Afterwards the trucks are loaded fully automatically. During retrieval mixed pallets can be created. This reduces the time needed for loading and helps to economise room for loading on the truck. The logistics software WAMAS by SSI Schaefer takes care of the administration, control and optimisation of the entire logistics process in the hub. 3,000 pallets leave the hub on a daily basis to be shipped all over the word. http://www.ssi-schaefer.com

Introduction to Vaccine Carriers and Cold Boxes Cold boxes, vaccine carriers and coolant packs are used for keeping vaccines cold during transportation. In comparison with cold boxes, vaccine carriers have a smaller volume, suitable for use by health workers during immunization campaigns and out-reach services. As these are passive devices, coolant packs are accessories for both cold boxes and vaccine carriers providing the cooling energy/warming for a limited time period. The products are, in principle, fairly simple. Overall, there is a low level of skills required to use them. Coolant packs are put into the boxes/carriers packed with the vaccine vials, in accordance with the instructions provided with the product (drawings inside the cover of the box/carrier). It is always recommended to order a second set of coolant packs for every cold box/ vaccine carrier, to be readily available for an interrupted service delivery at all levels. Cold boxes are insulated containers that can be lined with coolant packs to keep vaccines and diluents cold during transportation and/or short period storage. Cold boxes are used to collect and transport vaccine supplies from one fixed vaccine store to another, and from vaccine stores to health facilities. They are also used to temporarily store vaccines when the refrigerator is out of order or being defrosted. The vaccine storage capacity of cold boxes are between 5.0 and 25.0 litres. There are two types of cold boxes:  Short range: With a minimum cold life of 48 hours.  Long range: With a minimum cold life of 96 hours. Vaccine carriers are insulated containers that, when lined with coolant packs, keep vaccines and diluents cold during transportation. They are smaller than cold boxes and are easier to carry when walking. They are used for transporting vaccines from health facilities with refrigeration to outreach sessions where refrigeration and ice are not available. They are typically carried by a single health worker travelling on foot or by other means, where the combined journey time and immunization activity ranges from a few hours to a whole day. The vaccine storage capacity of vaccine carriers are between 0.1 and 5.0 litres. ________________________________________________________ 2. Terminology Cold life: The empty container is stabilized at +43°C and loaded with icepacks. Cold life is measured from the moment when the container lid is closed until the temperature of the warmest point in the vaccine storage compartment first reaches +10°C, at a constant ambient temperature of +43°C. Cool life: The empty container is stabilized at +43°C and loaded with coolant packs which have been stabilized at + 5°C for a minimum of 24 hours. Cool life is measured from the moment when the container is closed, until the temperature of the warmest point inside the vaccine storage compartment first reaches +20°C, at a constant ambient temperature of +43°C. Cool-pack: A water-pack pre-cooled to a temperature between + 2°C to +8°C before use. Ice-pack: A water-pack frozen to a temperature between -5°C and -20°C before use. Ice-packs are used frozen for the transport of oral polio vaccine (OPV), and used conditioned (see procedure for conditioning further down) for the transport of all other vaccines. Temperature Stability: The vaccine carriers and cold boxes are tested for temperature stability at ambient temperatures of 43°C and -20°C in WHO-accredited laboratories to ensure that at field conditions the equipment will perform optimally. The frequency and duration of door openings will raise the internal temperature of the boxes, and will shorten the Cold life and Cool life. _________________________________________________________ 3.
